沒(méi)食子酸對(duì)金黃色葡萄球菌抑菌活性及機(jī)制研究

【摘要】 以金黃色葡萄球菌為供試菌,利用2,3,5-氯化三苯基四氮唑(TTC)染色,細(xì)胞膜滲透性測(cè)定,SDS-PAGE蛋白譜變化等方法對(duì)沒(méi)食子酸的抑菌活性及其機(jī)制進(jìn)行研究,進(jìn)而驗(yàn)證沒(méi)食子酸的藥效作用。實(shí)驗(yàn)結(jié)果顯示,沒(méi)食子酸對(duì)金黃色葡萄球菌作用其抑菌圈直徑為13mm;其對(duì)金黃色葡萄球菌的最低抑菌濃度為0.01mg.mL-1;當(dāng)藥物作用5h時(shí),實(shí)驗(yàn)組的電導(dǎo)率與對(duì)照組相比增加了6.38%;大分子的OD值隨著作用時(shí)間的增加而逐漸增大,當(dāng)培養(yǎng)至8h時(shí),實(shí)驗(yàn)組大分子物質(zhì)吸光值的變化率達(dá)到最大,比對(duì)照組增加了74.4%。SDS-PAGE蛋白譜變化實(shí)驗(yàn)表明,沒(méi)食子酸對(duì)金黃色葡萄球菌作用20h后,其總蛋白量沒(méi)有明顯變化。上述結(jié)果表明,沒(méi)食子酸對(duì)金黃色葡萄球菌有較強(qiáng)的抑菌活性,能影響金黃色葡萄球菌細(xì)胞膜的通透性。其抑菌機(jī)制是通過(guò)破壞細(xì)菌細(xì)胞膜的完整性來(lái)實(shí)現(xiàn)的。
【Abstract】 To investigate the Antibacterial Activity and Mechanism of Gallic acid on Staphylococcus aureus.The antibacterial activity of Gallic acid was carried out by utilizing oxford cup and 2,3,5-triphenyl tetrazolium chloride(TTC),and the mechanisms of Gallic acid on Staphylococcus aureus were systematically investigated using determining membrane penetrability,the change of SDS-PAGE protein spectra and so on.It is found that Gallic acid has antibacterial activity on Staphylococcus aureus,and the minim...
